《古镇情缘》描述的是一场发生在珠三角一座城市、围绕祖屋的存与没而引发的爱情故事。影片以一个年轻华侨回到离开了二十多年的故乡为引子，牵扯出他与三位女主角之间或缠绵或无奈的爱恨纠葛。伊能静扮演女园艺师叶贝，将和另外两位女孩上演复杂的“夺夫大战”。 男主人公杨钧少年时随家人到秘鲁生活，转眼三十多岁了，事业生活多不如意。杨均回到故乡打算卖掉祖屋以图再起，不料故土情怀促使他改变初衷，做出留在家乡的决定。已长大成人的邻家女孩园艺师叶贝对这位儿时的大哥哥一见倾心，他也为叶贝浑身洋溢的青春气息所吸引，双双坠入爱河；而他的初恋情人、现已成为灯饰集团老总的欧妙娟婚姻破裂、感情空寂，面对昔日恋人一心想再续前缘。两个女人于是展开激烈的争夺大战。更有趣的是，一个火热的秘鲁女孩万里追赶，加入搅局，使这场情感戏变得更加扑朔迷离，云团万象。处在漩涡中的杨钧如何选择，戏如何收场……
